چکیده

Abstract The nutrient-sensor protein Sirtuin 1 (Sirt1; silent mating type information regulation 2 homolog 1) has been shown to have significant and opposing effects on insulin resistance, leptin body weight in the periphery brain. In hypothalamic arcuate nucleus (ARC) of brain, Sirt1 increases obese state acts promote gain as well resistance by increasing orexigenic neuropeptides Agouti-related (AgRP) neuropeptide Y (NPY), a distinct set ARC neurons, decreasing POMC thus its anorexigenic derivative alpha-melanocyte stimulating hormone (alpha-MSH) (1). Sirt1’s actions these are mediated at least part deacetylation transcription factor forkhead box O1 (FOXO1). Another mechanism which regulates appears be through mediating changes synapses neuropeptide-producing neurons. For example, previous study demonstrated that inhibition with specific inhibitor, Ex-527, decreased AgRP-NPY inhibitory synaptic input suggests obesity-induced increase would neurons promoting (2). present investigated how specifically POMC-producing N43-5 Results reveal Ex-527 significantly increased presynaptic marker Synapsin (Syn1) Furthermore, we whether target, FOXO1, mediates changes. FOXO1 overexpression Syn1 transfection mutant Syn1. Overall, our results suggest does so manner differs from neuronal synapses. Future work will elucidate mechanisms consequences neuron under different nutritional conditions vitro vivo. (1) Cyr, N. E., Steger, J. S., Toorie, A. M., Yang, Z., Stuart, R., Nillni, E. (2014). PDK-1/FoxO1 pathway in POMC neurons regulates Pomc expression and food intake.

Both insulin and leptin signaling converge on phosphatidylinositol 3-OH kinase [PI(3)K]/3-phosphoinositide-dependent protein kinase-1 (PDK-1)/protein kinase B (PKB, also known as Akt) in proopiomelanocortin (POMC) neurons. Forkhead box-containing protein-O1 (FoxO1) is inactivated in a PI(3)K-dependent manner. However, the interrelationship between PI(3)K/PDK-1/Akt and FoxO1, and the chronic eff...

Regulation of autophagy by AMP-activated protein kinase/ sirtuin 1 pathway reduces spinal cord neurons damage

Objective(s): AMP-activated protein kinase/sirtuin 1 (AMPK/SIRT1) signaling pathway has been proved to be involved in the regulation of autophagy in various models. The aim of this study was to evaluate the effect of AMPK/SIRT1 pathway on autophagy after spinal cord injury (SCI). Materials and Methods:The SCI model was established in rats in vivo and the primary spinal cord neurons were subject...

Glutaredoxin regulates vascular development by reversible glutathionylation of sirtuin 1.

Embryonic development depends on complex and precisely orchestrated signaling pathways including specific reduction/oxidation cascades. Oxidoreductases of the thioredoxin family are key players conveying redox signals through reversible posttranslational modifications of protein thiols. The importance of this protein family during embryogenesis has recently been exemplified for glutaredoxin 2, ...

Sirtuin 1 regulates lipid metabolism associated with optic nerve regeneration.

Injury to the optic nerve may lead to axonal degeneration, followed by the gradual death of retinal ganglion cells (RGCs), which results in irreversible vision loss. In the present study the mechanism of optic nerve injury, and the following regeneration and repair processes associated with sirtuin 1 (SIRT1)‑regulated lipid metabolism were analyzed. In addition, the treatment of optic nerve inj...

Blunted Refeeding Response and Increased Locomotor Activity in Mice Lacking FoxO1 in Synapsin-Cre–Expressing Neurons

Successful development of antiobesity agents requires detailed knowledge of neural pathways controlling body weight, eating behavior, and peripheral metabolism. Genetic ablation of FoxO1 in selected hypothalamic neurons decreases food intake, increases energy expenditure, and improves glucose homeostasis, highlighting the role of this gene in insulin and leptin signaling. However, little is kno...
